ClavystBio Conversations with Robert Abraham

ClavystBio hosted Robert (Bob) Abraham, Chief Scientific Officer of Engine Biosciences, for a fireside chat with Amy Schulman, Managing Partner at Polaris Partners.

Drawing on decades of experience in drug discovery and development, Bob shared his insights into the role of AI in understanding disease biology, the challenges of drug development, and the journey from scientific discovery to new medicines. Here are some key insights from the discussion:

  1. AI’s Strength Is Making Sense of Complex Disease Biology
AI’s value lies in finding patterns across large, diverse datasets to understand diseases that are difficult to explain through a single biological hypothesis. Examples include polygenic mutations, protein expressions and ageing-related diseases.
  2. Academia and Startups Carry Different Expectations
There is no hard and fast rule on the role of scientific founders after a spin-off. The key is to ensure expectations are clear.
  3. Defining Patient Population for Clinical Trials Is a Critical Task
Restricting patient enrolment based on very specific biomarkers may give better efficacy signals, but may also increase enrolment time and costs, and artificially constrain the patient population. It is important to be certain before making definitive decisions.
  4. Curiosity-Driven Research Lays the Groundwork for Future Medicines
Many important therapies began with basic research that had no obvious near-term application. For example, palbociclib began with cell cycle research, while CRISPR traced back to studies on bacteria and viral infections. Shifting too heavily towards translational research today could leave gaps in the pipeline that only become visible years later.

Amy also highlighted Singapore's strong research base and the opportunity to support translational efforts in building globally relevant companies. The discussion underscored the importance of connecting scientific discovery with commercial development to advance healthcare innovation.
